Calico, Green, Hairy

This page serves as a repository of research notes on Toxin RP13, the thirteenth mutagen unlocked in River Plains by Renard. It uses 4 Calico, Green, Hairy (Negative to Fuzzy, Mottled, Spongy) moss and 3 Eye of Osiris, 4 Falcon's Bait, 2 Hairy Tooth, 1 Nature Jug, 1 Scorpion Brood, 2 Sun Star mushrooms.

First mutation test: L-Vampire R-Silken

Magenta is missing from Vampire's outer stamen, no change is apparent on the Silken.

Second mutation test: L-Silken R-Vampire

Silken's outer stamen has changed to have yellow in only the outer stamen. Either a Yellow was transferred into the midst of the Is, or an I was transferred into the midst of the Ys. Let's go with the second assumption until a contradiction shows up.

Left-Silken 8(Y)

Vampire has lost yellow in the outer stamen.

Right-Vampire 1 or 7. (Not 3 because if so yellow would be visible on the entire stamen)

Discussion and Next Step

We know from the second test that the gene affected on Vampire on the left is either 5 or 6. If the gene on the right splint was gene 1 on Vampire, that would be genes 1 or 2 (R) on Silken. This is not supportable since either there would be blue in Silken's stamen (if an I was transferred from Vampire) or blue in Vampire's outer stamen (if an O were transferred). Hence,

Right-Vampire 7(I)

That narrows the range on Vampire and Silken a bit.

Third mutation test: L-Clarity R-Delicate

On the left, a yellow north was broken. That's part of the GRRO allele, far to the right of where the assumed range was. Time to reconsider previous work!

Fourth mutation test: L-Delicate R-Clarity

No change to either flower.

Reworking old assumptions

We've established now that the left splint hits somewhat right of center, at least gene 21 of 36. Therefore, in the second test, it must have been an I that was transferred to the Vampire and swapped with a Y. That puts us on the far right of the genome, the last two genes.

So what do we know about the right splint? Since a Y was transferred in the second test, the right splint is at or less than 66.7%. In test 1 when Vampire and Silken crossed, Vampire lost a color on the outer stamen and did not gain any color, while Silken did not gain any color. Vampire losing color could be explained by losing an I, or losing a color gene which wasn't replaced by a different color gene. If Silken on the right did not transfer a color gene, that would be gene 10 or above. However, an I might have been transferred to replace gene 9. Therefore the range on the right splint is somewhere between 44.4 - 66.7%. For Vampire on the right, that's genes 6, 7, or 8.

On the second test, an I was transferred to Vampire, replacing a Y. That has only one possibility.

Right-Vampire 8(Y)

We can now say that:
Left-Vampire: 11-12(IO) Right-Silken: 11-12(GG)

Let's reanalyze the possibilities for Delicate and Clarity.

Third mutation test: L-Clarity R-Delicate

On the right, a yellow North (GRRO) was broken, no change to the stamen. On the right, yellow was lost to the outer stamen.

Two possibilities. Either an I (Gene 16) was transferred from Delicate to the far right of Clarity, or a G was transferred from Clarity to replace gene 15 on Delicate.

Fourth mutation test: L-Delicate R-Clarity

No change to either flower.

Discussion

We'll recall from the third test that either a gene was transferred from the left side of the plausible range on the left-splint to the left side of the plausible range on the right-splint, or a right-right plausible range transfer. Left-left is implausible here, so I will act under the presumption that we're dealing with the following:
Left-Delicate 16(R)
Right-Clarity 23(R)

That limits the range of possibilities from the third test to:
Left-Clarity 36(O)
Right-Delicate 11(I)

Which leads to:
Left-Vampire 12(O)
Right-Silken 12(G)

The range on the right now is surprisingly narrow, and on the left it's quite specific. It might actually be a good time to test Crimson.

Crimson (Genes 103-106): GRRO
Left-Splint: 104-106
At first it seems like putting an I into this gene set would be informative, but Crimson is a difficult gene to test on the stamen. Colors won't appear on the stamen beyond four genes, so I wouldn't actually be able to tell the difference between putting an I on gene 105 and 106. However, swapping in an O or an R would be ambiguous on genes 104 and 105, so that's not any better. I'll do a cross with Clarity on the right anyways and donate an R. A hit on gene 106 will be clear, and a hit on genes 104 or 105 can be clarified with further testing.

Crimson ():
GYYY Right-Splint: 67-68
Crossing with Silken will donate an I, and results will be clear on the stamen.

Fifth mutation test: L-Crimson, R-Clarity

Both flowers are unchanged, the ambiguous result. Therefore, the result will be clear if I can add an I to the left splint. No change will be apparent on Crimson if an I is added to gene 105 since there are already four magenta on the outer stamen. However, if gene 104 is affected, the appearance on Cyan will be visible.

Sixth mutation test: L-Silken, R-Crimson

Color is visible on the middle stamen, but not the inner stamen.
Right-Crimson 68(Y)

Next step

The range on the right-splint is now so narrow, that the only flower without a fully characterized result is Morning.

Morning: URORGRRR
Right-splint: 28-29
This is trivial to test, any change will affect a different petal. In fact, we can do a Morning-Morning cross which might even help narrow down the left splint.

Seventh mutation test: L-Crimson, R-Delicate

No change to the outer stamen.
Left-Crimson 105(R)

Eighth mutation test: L-Morning, R-Morning

On the left, GRRO has become GRRR.
Left-Morning: 44(O)
Right-Morning: 28(R)

Final test

Everything has been characterized but Left-Dusk which is either 72 or 73.

Ninth mutation test: L-Dusk, R-Delicate

No change to the Dusk outer stamen.
Left-Dusk 73(O)
